Big daddies without little sisters.. HELP!

The only big daddies I killed I did get sisters from. These are two I didn't kill previously.
There aren't any little sister icons on the map. The pause menu shows 2 remain in the level, but I'm not finding any on the map. It's still playing that big scary music and telling me to get on the train, but I should still be able to get the little sister. If I get on the train and leave, these are lost forever, aren't they? Been following this one around in the diner area and this one big daddy keeps walking around to vents, knocking and nothing comes out, and he goes to another vent.
Oh, and yes, random splicers are still spawning everywhere.

Update: I killed both of the big daddies, not seeing anything. I went to both vents, even tried knocking on them with my drill. I created a second save file for this in case anyone has the solution to this issue.. PLEASE! The pause menu still shows that 2 little sisters remain in the room and the map shows no little sister icons. Is there a chance that I accidentally killed them earlier when I was going after splicers? Can they die, and do they re-spawn?

Not all Bid Daddies have Little Sisters.

There are only a limited number of them in each level.

The splash menu tells you the number.

The pause menu shows "little sisters in this level" and has 2 icons. I BELIEVE I've already collected three.. maybe only two and the game doesn't like strike out how many you've collected so far? Once you've defeated a big sister in an area, is it safe to assume that there aren't any other little sisters to collect?

big daddies respawn after a time, without sisters, they dont x them out but the icon changes to a little sister face

Heh.. so if it says 'little sisters in this level' and it shows TWO FACES, I've already collected them and I've been wasting my time like an idiot? Win!

No, use the pause menu for the amount of Little Sisters to guide you.

I don't remember if it updates the menu as you take them out, but I don't think it does, it just tells you the number of available Little Sisters on each level.

As far as I know, Big Sister spawns after you dealt with all Little Sisters on level.

It shows both number and "state" of Little Sisters on level:
- LS with Daddy Icon - not dealt with.
- Normal Girl - saved.
- LS with x's on her eyes and drop of blood comming out of mouth - harvested.
