No you cannot hurt little sisters in any way other than harvesting.
You can only kill them by Harvesting them, it will have a good explanation in the game Ken Levine said in the Uncle Gamer Interview a few months back.
You can't because if you misfired (incredibly easy to do, I don't care how good you are, it would happen), your choice to take the Adam from her or rescue her would be taken away.

Imo, they still couldve let you kill them with your weapons then harvest the ADAM from their dead body, i think though they have to be alive to extract the ADAM...it's just a smart spin on hopw to avoid really nasty violence and controversy with the media.
but what if you want to save them instead? having them accidentally killed all the time would be a huge pain in the rear end.
They would get killed in cross fire from Splicers too. It isn't just you that would be able to kill them.
Trust me, making them stay alive untill they are harvested is the only option that doesn't break the game.

Yes, the Splicers want them just as badly as you do. The differance is that a Splicer is AI, and would be able to kill a LS on the other side of Rapture without you being able to stop it.
The Splicer might not even want to kill the LS, it just missed and hit her, killing her. Once again, you wouldn't be able to do anything about it.
Trust me, it would suck to have the game any other way.
I was under the impressing that for you to interact in anyway the little sisters, you fist had to kill the big daddy, and then you would make your choice. You cant just shoot them while the big daddy is attacking you.
