Big Daddies are humans in special suits, volunteers who guard the Little Sisters.

So why/how do they make inhuman grunts and growls? They seem so monstrous, there's no way they can be humans.

You know what I find funny about this? Everyones gotta eat sleep dispense of "waste" etc. So, this either means that once they go daddy, there is no turning back. Considering everytime they have to take care of any personal business, they'd have to take it off, put it back on which seems like it'd be too tedius. So they have to have some kind of inhuman quality once they modify themselves to fit the Big Daddy role. I hope it will be cleared up in the sequel/expansion.
P.S. This game is amazing, seriously. :D

Damn, I wish they were monsters, that'd make it more interesting. It makes me sad that they're just humans, though.

I have been working on compiling an article called "Analyzing the Big Daddy" based on information provided in the game, scenarios presented in the trailers, and whatever tidbits I can pull off the internet that sound feasible. I'll try to keep this short though (and of course, I'm not saying that my word is the final word on anything, simply what I believe).
PLEASE KEEP IN MIND, BIG SPOILERS BELOW, DO NOT READ IF YOU DO NOT WANT TO HAVE THE GAME SPOILED. HIGHLIGHT THE AREA BETWEEN THE THREE DASHES IF YOU STILL WANT TO READ.

I think that there are two "levels" of becoming a Big Daddy. The first I simply refer to as being a "partial transformation". An example of this would be Jack: he got his larynx (voicebox) modified via a machine, he wore the whole suit, and he injected himself with pheromones, but he still retained his human thought process and emotions. He was not constrained to a Little Sister; you could walk off and let her die if you wanted. Granted you would have to go and get another one to progress, but you never see any Big Daddies ingame that would leave their Little Sister to die. He also did not seem to modify his body in any other way; he does not walk with a slow pace, and he retains use of all his Plasmids and Weapons. And, in the end, he is able to take off his diving suit and return to the surface to lead the rest of his live as normal, so it is clear that he had not been drastically modified. So, partial transformation.
The Big Daddies that you fight, however, are much more than this, and much less at the same time. Their bodies are huge and deformed, and have been described even as being "biomechanical". True, they may have at one point been human volunteers, but science has warped them far past that point. I doubt they could remove their suit and be recognizable or acceptable up on the surface-- to a human, they would probably indeed look incredibly alien. Another point is behavior... they exist only to protect their Little Sister. The Big Daddies without Little Sisters will simply plod around Rapture pointlessly, banging on any vents that they come to. It seems that the Bouncer have even been modified in some way that allows them to run even faster than a human when pushed into "aggressive mode". Many of these traits that are present in the enemy Big Daddies are not present in Jack when he is "turned". So yes, you could say they are human, but in reality there is very little left of them that actually is "human".

Anyways, that's my basic take on it. I'm not trying to be high and mighty here-- like I said, I am by no means the final word. I simply think it is fun to analyze fictional things and try to make them seem feasible.

It's strange they say it's irreversible, must have been some propaganda they fed the volunteers since in the end you are human again watching the cinema. So at least one person reversed it.

Fontain's Home for the poor - i.e. suckers we can turn into Big Daddies.
Little Sister Orphanage - i.e. Kids we can steal to turn into Adam production factories.
It's refreshing to know that not every game has to have some sort of monster in it. That somebody could take the human form and make it more intimidating by giving it a few modifications. Not everything has to be about some sort of monster, after all aren't humans the greatest monsters of all?

The Big Daddies are humans who voluntered to become the protectors of the little sisters turning them into genetically modiffied monster. note: look for audio diaries near games end (you will know when)
