From a film or game it's easily pacino, deniro or pesci. But I have to take my hat off to chazz palminteri, he did a brilliant job with A Bronx Tale. One of my favourite movies of all time.
I know you didn't ask for real life gangsters but I have nothing but respect for Jackie the Nose (John D'Amico), Louis Ricco, Lucky Luciano and a few others.

What Michael Corleone did in the end of Godfather 1 was probably one of the most badass things I've seen in a Mafia movie, so he's my favourite so far :D

Is it favorite Mafia actor or Mafia character?
My favorite Mafia actor, hmm tough one i'd say it is a tie between Gandofini, Liotta and Imperioli.
Favorite Mafia Character would be probably Tony S.
My least favorite (just for fun) would be Phil Leotardo... and Billy Bats. Both portrayed by the awesome Frank Vincent.

Favourite character:
(you'll probably think I'm joking but) Fat Tony from the Simpsons
or maybe Tony Montana or (and again you'll probably think I'm joking) Michael Corleone as dipicted in Godfather 3
(As a rule I don't watch Gangster Films, I'm more into love stories as long as they're not overtly chick-flick)
Favourite actual, real gangster...
Pablo Escobar, because when the government got too close to his operations he hired a private army and tried to bulldoze the government buildings. That basically just appeals to my sense of humour :D

Well, since so many others are going with Pacino as Michael Corleone, I'll dare to be different & take a more a classical approach. :)
James Cagney. He was the guy that real life gangsters used to try & imitate back in the day. Cagney was only about 5'6", but he carried himself on screen like a giant.
A few of Cagney's best:
The Public Enemy (http://www.imdb.com/title/tt0022286/)
Angels With Dirty Faces (http://www.imdb.com/title/tt0029870/)
White Heat (http://www.imdb.com/title/tt0042041/)

Real Life:
Dutch Schultz. Likely the most violent of his contemporaries. He had a notorious temper and was not afraid to get his hands dirty himself, as exemplified in his friend's Dixie Davis' account of his execution of Julie Martin. I've read accounts that he alledgedly had more than 100 people whacked over a 5 year period. Really, everything about Schultz justs screams toughness and machismo (except possibly his real name) to a level most other gangsters can only dream of. Furthermore, there is still the legend of his buried treasure.
Film:
Tom Reagan. Undoubtedly the smartest gangster ever put to film, Reagan is constantly two steps ahead of everyone else. That is, until the Dane takes issue with his story and drags him out for a walk in the woods to make sure he really did kill Bernie, which to my mind is the greatest scene in "Miller's Crossing". Outside of that scene, Reagan is always in control and the puppetmaster who skillfully manipulates events in favour of his boss.
